1) Measure 1.00g Salmon Sperm DNA (Sigma# D-1626)

2) Dissolve in 100 ml Milli-Q Water using magnetic stirrer

-This may take overnight at room temp

-Cover with parafilm to prevent evaporation

3) Shear DNA by passing several times through 18-20 guage

hypodermic needle

4) Boil for 10 minutes

6) Store at -20'C
