Transformation into XL1-Blue Competent cells (Stratagene 200249)

1. Prechil 14ml polypropylene round-bottom tubes on ice, and preheat SOC to 42°C

2. Thaw cells on ice, put 100µl cells into each tube

3. Add 1.7µl ßme to each tube

4. Swirl gently every 2 minutes for 10 minutes

5. Add 0.1 to 50ng DNA to each tube, swirl to mix

6. Incubate 30 minutes on ice

7. Heat shock 42°C for exactly 45 seconds

8. Incubate on ice 2 minutes

9. Add 0.9ml pre-heated SOC (42°C) to cells and incubate 37°C for 1 hour

10. Plate <200µl per plate

11. Incubate plate overnight 37°C

Total time just under 2 hours
